氟对体外培养软骨生长和凋亡的影响及SOD对氟的拮抗作用
Influence of fluoride on the growth and apoptosis of cultured cartilage and antagonizing effect of su- peroxkte dismutase (SOD)
【摘要】 目的 研究氟对体外培养软骨生长和凋亡的影响以及SOD对氟的拮抗作用。方法 取兔肋软骨进行体外培养,在培养液中加入不同浓度的NaF,以及NaF+SOD,采用图像分析仪检测软骨细胞生长的情况;用原位末端标记检测技术检测细胞凋亡。结果 与对照组相比,NaF 20μmol/L、NaF320μmol/L使软骨细胞柱明显增长,面积、周长增加,直径减少;NaF 320μmol/L可使肥大软骨细胞显著凋亡;NaF320μmol/L+SOD则使软骨细胞柱长度明显减少,面积、周长减少;与不加SOD的同浓度NaF组相比,NaF20μmol/L+SOD、NaF320μmol/L+SOD组肥大软骨细胞凋亡明显减少。结论 (1)中、低剂量的氟可引起软骨佝偻病样损害,即肥大软骨细胞明显增多,肥大软骨细胞发生大量凋亡。SOD可以阻断上述改变。(2)中、低剂量的氟引起软骨的病变可能与刺激软骨细胞产生多量自由基,从而引起大量软骨细胞凋亡有关。
【Abstract】 Objective To study the influence of fluoride on the growth and apoptosis of cultured cartilage and the antagonizing effect of SOD. Methods The cartilage from the ribs of newborn rabbit had been cultured in vitro for 18 days. Various concentration fluoride with or without SOD was added to the culture medium. Apoptosis was observed by terminal deoxyribonucleotide transferase-mediated dUTP-bintin (TUNED method. The morphological changes were assayed by image analysis. Results(1) Apoptosis was obviously increased at 320μmol/L of NaF. The height, area and circumference of the cartilage cell column were increased while the diameter decreased at concentration of 20μmol/L and 320μmol/L. (2) When SOD was added the morphological changes mentioned above could not observed at 320μmol/L, and the count of apoptosis decreased at 20μmol/L and 320μmol/L. Conclusions (1) The results suggested that fluoride at low and medial concentration could induce apoptosis and proliferation of the hypertrophic chondrocytes. SOD could protect the cells from the damage caused by fluoride.(2) Cell apoptosis induced by free radicals hence, which was promoted producing by fluoride at low and medial concentration, might be the reason of the cartilage damage.
