大鼠骨髓间充质干细胞体内向造血细胞分化的研究
Preliminary Study on Hematopoietic Differentiation Potential after Culture-Expanded Rat MSCs Transplantation in BALB/C Mice
【摘要】 目的:研究大鼠骨髓间充质干细胞(MSC)及其天麻定向诱导神经细胞后向造血细胞分化的潜能。 方法:建立以亚致死量照射的BALB/C小鼠为受体动物模型,体外扩增的大鼠6~7代MSC为供体细胞,分以 下3组,每组12只进行实验:实验组1(A组),放疗+单纯输注MSC;实验组2(B组),放疗+输注天麻诱导1h MSC,A、B组每只小鼠注射0.3mL含1.5×106cells的无血清培养液。空白对照组,放疗+输注等量无血清培 养液。照射后4h内,分别经尾静脉注射。移植60d后取骨髓(BM)、外周血(PB)、脾(SP)细胞悬液,用流式细 胞仪检测供体大鼠源性CD11a、CD45表达。结果:所有实验小鼠均能存活到2个月,骨髓、外周血、脾细胞悬液 均可检测到低表达的大鼠源性CD11a、CD45。对照组为阴性。A、B两组相互比较,具有显著差别意义(P< 0.05)。结论:MSC及天麻定向诱导神经细胞后具有向造血细胞分化潜能。
【Abstract】 Objective:To investigate the differentiation potential of rat mesenchymal stem cells(MSCs) and the neurons derived from MSCs induced by Gastrodin into hematopoietic cells in vivo. Methods:Donor cells were derived from rat MSCs of passage 6~7, and BALB/C mice pretreated with sublethal irradiation provided an in-vivo transplantation model.There had 2 experimental groups:MSCs transplantation group(group 1),MSCs induced into neurons by Gastrodia elata group(group 2) and control group(pretreated with sublethal irradiation and transplanted with DMEM-LG). The expression of rat CD11a and CD45 were studied by FCM on the 60th day after transplantation. Result:All balb/c mice survived within 60 days. Low level of rat MSCs-derived CD11a and CD45 could be detected by FCM in bone marrow(BM), peripheral blood(PB), and spleen(SP) cells, But cannot detected in the control group.The percentage of the donor cell-derived CD11a and CD45+ cells in Group 2 was significantly higher than that of group 1(P<0.05). Conclusion:MSCs and neurons derived from MSCs induced by Gastrodia elata have ability to differentiate into hematopoietic stem cells/ progenitor cells.
