冷冻对人类成熟及未成熟卵发展潜能的影响

【摘要】 目的:探索冷冻卵子的成熟时期。方法:将153个生发泡期(GV期)人类卵子分为3组,对照组:体外培养至成熟(MⅡ期),行单精子胞浆内注射(ICSI),MⅡ期冷冻组:体外成熟至MⅡ,然后将MⅡ期卵子进行冷冻,解冻后将存活的卵子行ICSI;GV期冷冻组:将GV期卵子进行冷冻,解冻后体外培养至MⅡ,然后再行ICSI。用慢速冷冻快速解冻的方法对成熟的和未成熟的卵子进行冷冻和解冻,观察体外成熟情况、解冻后的存活情况、受精情况及胚胎发育情况。结果:体外成熟率:GV期冷冻组51.2%,对照组为70.0%,有显著差异;受精率:3组分别为68.0%、33.3%、42.1%,有显著差异。对照组的发展潜能要显著优于试验组;GV期冷冻组的存活率、受精率优于MⅡ期冷冻组。结论:冷冻会对卵子的发育潜能有1定的损伤,在GV期进行冷冻可降低这种伤害。

【Abstract】 Objective: To probe the mature stage of cryopreservation human oocytes. Methods: 153 human oocytes in GV stage were divided into three groups, control group, performing ICSI after culturing to M Ⅱ in vitro; M Ⅱ frozen group, maturating the GV stage oocytes to M Ⅱ in vitro, freezing them subsequently and performing ICSI after thawing; GV frozen group, freezing GV-stage oocytes, then culturing them to M Ⅱ in vitro after thawing, selecting the survivals for ICSI. To compare the maturation rate, survival rate, insemination rate and embryo cleavage rate. Results: maturation rate in vitru: GV frozen group 51.2%, control group 70.0%; insemination rate: control group68.0%, GV frozen group 42.1%, M Ⅱ frozen group 33.3%. the development potential of control group is prior to that of the either trial group. The survival rate, insemination rate of GV frozen group are higher than those of M Ⅱ frozen group. Conclusion: oocytes frozen in GV stage can decrease the damages on development potential.
