【摘要】 目的 检测尤文肉瘤患者外周血单个核细胞中的EWS/FLI-1mRNA的表达并且探讨其临床意义。方法 应用巢式RT-PCR的方法检测11例尤文肉瘤患者15份标本和20例健康者外周血标本中EWS/FLI-1mRNA的表达。结果 全部健康志愿者外周血标本中均没有发现EWS/FLI-1mRNA的表达。11例尤文肉瘤患者的15份标本中,9例病人(81.8％)的13份标本中检测到有EWS/FLI-1mRNA的表达。其中6例病人(66.7％)在随访中发生转移,其EWS/FLI-1mRNA的表达水平明显高于另外3例没有发生转移的病例(P<0.05)。结论 应用巢式RT-PCR方法检测尤文肉瘤患者外周血中EWS/FLI-1mRNA的表达是一种帮助判断预后和评估治疗效果的比较好的方法。更多还原

【Abstract】 Objective To detect the expression of EWS/SLI - 1mRNA in the peripheral blood mononu-clear cells of patients with Ewing’ s sarcoma and assess the clinical significance. Methods Peripheral blood EWS/SLI - 1 mRNA expression was detected by means of RT - nested PCR in 11 patients with Ewing’ s sarcoma and 20 healthy subjects. Results No EWS/SLI - ImRNA was detected in all 20 ( 100% ) healthy subjects; on the contrary, positive results were observed in 13 peripheral blood samples of 9 (66. 7% ) patients among 15 peripheral blood samples of 11 patients with Ewing’ s sarcoma. Six of these patients subsequently developed clinical metastasis, the level of EWS/SLI - 1mRNA expression of these 6 patients were much higher than that of the other 3 patients whom there was no evidence of metastasis ( P < 0. 05) . Conclusions Detection of the expression of EWS/SLI - 1mRNA in the peripheral blood mononuclear cells of patients with Ewing’ s sarcoma by means of RT -nested - PCR is an acceptable method to predict the prognosis and assess the results of therapy.更多还原
