微乳液法制备纳米二氧化硅基因传递复合载体

【摘要】 以硅酸盐溶液/环己烷/聚乙二醇辛基苯基醚/正戊醇体系的微乳液为原料反应,以浓硫酸为沉淀剂,采用微乳液法制备出了高比表面积纳米SiO2粉体,由BET,TEM,IR,XRD和DTATG对样品进行了形貌表征和物相分析。用多聚赖氨酸在纳米SiO2颗粒进行表面修饰,制得多聚赖氨酸/SiO2复合载体材料。结果表明:采用微乳液法制备的纳米SiO2粉末均由非晶态物质组成,粉体颗粒形貌呈近似球形,粒径为15~35nm,比表面积高达580~630m2/g。通过脱氧核糖核酸(deoxyribonucleicacid,DNA)结合及细胞转染实验可知,该纳米复合颗粒能高效传递DNA进入鼻咽癌细胞株(humannasopharyngealepithelioma1,HNE1)内,并产生较好的绿色荧光蛋白。

【Abstract】 Nano-sized SiO2 particles with high specific surface area were prepared by microemulsion reaction method using the microemulsion system of silicate solution, cyclohexane, OP-10(polyethylene glycol octylphenyl ether), and n-pentanol as raw materials, and concentrated sulfuric acid as precipitant. The prepared powders were characterized by means of BET (BrunauerEnmetTeller), TEM, IR, XRD and DTATG. Poly-L-lysine/SiO2composite carrier material was prepared by modifying SiO2 nanoparticles with poly-L-lysine. The results show that nano-sized SiO2 powders obtained by microemulsion method are amorphous powders in approximate spherical shape, their grains size is 15-35nm and the specific surface area of powders is up to 580-630 m2/g. Through the complexes of particles with plasmid deoxyribonucleic acid (DNA) and the cell transfect test, such composite nanoparticles can transfer DNA into human nasopharyngeal epithelioma 1( HNE1) cell efficiently and get good green fluorescent protein expression.
