【摘要】 以聚乙二醇(PEG)1000 为相转移催化剂,负载型杂多酸为催化剂,在乙酸介质中,用双氧水氧化 3,4,5-三甲氧基甲苯制备了合成辅酶 Q10的重要中间体 2,3-二甲氧基-5-甲基-1,4-苯醌,也称辅酶 Q0。研究获得了较合适的反应条件:反应温度在 30~35℃,24g 3,4,5-三甲氧基甲苯原料需要 0.4g PEG-1000 催化剂,乙酸和 50%双氧水各 40mL,20mL 负载型杂多酸催化剂。反应液经过水洗、萃取和结晶提纯后,辅酶 Q0 纯度达到 97%,收率为 90%。

【Abstract】 2,3-Dimethoxy-5-methyl-1,4-benzoquinone called coenzyme Q0,a key fragment in coenzyme Qn series and a key intermediate for synthesizing coenzyme Q10 was prepared from 3,4,5-trimethoxy toluene using polyethylene glycol (PEG-1000) as phase transfer catalyst(PTC), as well as supported heteropolyacid as acid catalyst, acetic acid as medium, hydrogen peroxide as oxidant. The suitable condition was found that 40mL acetic acid, 40mL 50% hydrogen peroxide, 20mL supported heteropolyacid catalyst and 0.4g PEG-1000 are required for 24g raw material 3,4,5-trimethoxy-toluene and the reacting temperature is 30~35 ℃. Via washing reaction medium with water, extraction with dichloromethane and removal of solvent, the crystal of product was prepared with the purity and yield of 97% and 90% respectively.
