微波消解石墨炉原子吸收法测定大鼠组织中的硒

【摘要】 由于易挥发元素硒在样品消解过程及含量测定的灰化阶段会有严重损失,为了解决硒的挥发损失及基体干扰等问题,采用无机钯为基体改进剂,微波消解—石墨炉原子吸收法对亚慢性汞硒中毒大鼠体内十种组织器官中硒的含量分别进行了测定与分析。研究结果表明,实验最佳用量的消化试剂为4mL的(10∶3)HNO3H2O2,最佳用量的基体改进剂为50μg·mL-1的氯化钯,而石墨炉升温程序中的最佳灰化温度和原子化温度分别为1200和1800℃。在以上最佳实验条件的前提下,线性测定范围为0~80ng·mL-1,检出限为183ng·mL-1,相对标准偏差小于8%,平均回收率为976%。由此可以看出,测定的方法准确、可靠、简便、快速,能够适用于生物体内多种组织器官中硒含量的测定与分析。

【Abstract】 The content of selenium in rat′s ten organs was determined by graphite furnace atomic absorption spectrometry with microwave dissolution. Inorganic palladium modifier was added into the sample to solve the problems of selenium volatilization and matrix disturbance as selenium in the sample is liable to receive serious loss during the course of digestion and cineration. The optimistic determination conditions were shown as follow, digestion reagent of 4 mL (10∶3)HNO 3/H 2O 2, matrix modification of 50 μg·mL -1 palladium chloride, cineration temperature of 1 200 ℃ and atomization temperature of 1 800 ℃. Under such optimum conditions, the linear range is 0 80 ng·mL -1 and the detection limit is 1 83 ng·mL -1 . The RSD is less than 8% and the average recovery rate is 97 6%. The method was shown to be precise, reliable, convenient and quick in selenium determination of various organism organs.
