桃果实中花青苷的提取、检测及应用

【摘要】 采用0.05%盐酸-甲醇提取液4℃浸取组织24h,可以高效提取桃果实果皮中的花青苷,方法回收率可达94.37%;采用梯度洗脱HPLC法,可以对花青苷进行定性、定量分析,该体系检测限(S/N=3)为0.2μg/g,RSD(n=5)为2.94%,峰面积与浓度在0.0625~0.2500mg/mL范围内线性关系达到极显著水平(r=0.997)。方法简便易行,可用于桃果实等材料的花青苷水平变化分析。

【Abstract】 Using peach fruit as material we established an effective extr action and analysis system.Anthocyanins could be efficiently extracted from pea ch epidermis with0.05%HCl-methanol solution for24h at4℃.Gradient elu tion was applied in HPLC analysis,and the recovery was found to be as high as9 4.37%,and the limit of determination(S/N=3)was0.2ìg/g.There was a significantly positive correlation between HPLC peak area and anthocyanin con centration ranging from0.0625to0.25mg/mL,with a correlative coeffic ient of0.997and a RSD of2.94%.The system was simple,feasible an d can be used to analyze anthocyanin in peach fruit.
