兔晶状体皮质吸出后FGF-R1mRNA在细胞增殖中的作用

【摘要】 目的:探讨碱性成纤维细胞生长因子受体1(fi-broblastgrowthfactorreceptor1,FGF-R1)在兔晶状体上皮细胞增殖中的作用。方法:将健康白色家兔42只随机分入实验组(36只)和对照组(6只)。实验组兔行双眼透明晶状体皮质吸除术,对照组未做处理。在术后1,3,7,14,30,60d各处死6只。应用免疫组化和RT—PCR方法检测赤道部晶状体上皮细胞中增殖细胞核抗原(proliferatingcellnuclearantigen,PCNA)及FGF-R1mRNA的表达。结果:FGF-R1mRNA相对含量随术后时间的发展,表现出明显的变化。术后1~3d时FGF-R1表达迅速升高到最高值。术后7d时仍维持在较高水平状态。14d后明显下降,逐渐恢复术前状态。FGF-R1mRNA与PCNA的表达呈正相关。结论:FGF-R1参与兔晶状体上皮细胞的增殖代谢过程,与细胞的增殖状态相关。

【Abstract】 AIM: To assess the effects of FGF-R1 mRNA on the proliferation of rabbit lens epithelial cells (LECs). · METHODS: A total of 42 white healthy rabbits were randomly divided into Exper imental Group (36) and Control Group (6). Rabbits in Experimental Group underwen t binocular lens cortex aspiration, while those in Control Group not. Six rabbi ts were killed at 1,3,7,14,30,60d after the surgery respectively. The expression s of proliferation cell nuclear antigen (PCNA) and FGF-R1 mRNA in rabbit LECs in different periods after lens cortex absorption were detected by immuno-histoche mical and RT-PCR methods respectively. · RESULTS: The expression of FGF-R1 mRNA was strongest within 1-3d after the su rgery, remaining on the high level at 7d postoperatively, and it noticeably decr eased at 14d after the surgery. The expression of FGF-R1 mRNA restored to its pr eoperative level till postoperative 1-2mo. · CONCLUSION: The expression of FGF-R1 mRNA is present in the normal rabbit epi thelial cells, and FGF-R1 mRNA participates in the procedure of proliferation of rabbit lens epithelial cells. ·
