【摘要】 目的:晶状体上皮细胞产生纤维生长因子并表达纤维生长因子的受体。本实验的研究目的是调查和研究晶状体上皮细胞增生调节和发育的规律。方法:通过使用牛晶状体上皮细胞培养后,使用“创口愈合方法”检查晶状体上皮细胞的增生和移动,用刮胡刀片在培养的牛晶状体上皮细胞上做一单层晶状体上皮细胞,然后加入一定介质观察和测量晶状体上皮细胞的数量、DNA合成及PCR方法检测基因产量。结果:suramin, protamine和抗纤维生长因子抗体抑制晶状体上皮细胞移动,晶状体上皮细胞的DNA合成也受到了抑制。结论:晶状体组织存在着纤维生长因子的调节系统,而且,这种调节系统在晶状体上皮细胞的增生和分化中起到重要的作用。

【Abstract】 AIM: To study the regulatory mechanism of LE (lens epithelial cell) proliferation and growth.· METHODS: Cultured bovine LEs (1-3rd passage) were used. In vitro "Wound Healing Method" was used to examine the proliferation and migration of LEs. Monolayer cells were wounded with a razor blade and the number of migrating cells was counted. Polyrase chain reaction (PCR) was performed to detect the gene product from LEs.· RESULTS: Migration of LEs into the open space of a culture dish was inhibited with suramin, protamine and anti-b-FGF antibody. DNA synthesis of LEs was also suppressed with these reagents.· CONCLUSION: In lens tissue, there is a regulating system for b-FGF (basic fibroblast growth factor), which might be important in the proliferation and differentiation of LEs.·
