碱性成纤维细胞生长因子对视网膜组织中一氧化氮及其合成酶的影响

【摘要】 目的 :观察碱性成纤维细胞生长因子 (b FGF)对成年家兔体外培养的视网膜组织的一氧化氮 (NO)及其合成酶 (NOS)的影响 ,旨在为视网膜移植提供抗氧化的保护方法。方法 :健康成年家兔 2 0只 (40眼 ) ,分成 4组 , (新鲜视网膜组 )、 (常规培养液组 )、 (高浓度b FGF液组 )、 (低浓度 b FGF液组 ) ,将 ~ 组视网膜组织块放入相应培养基中培养 1周。 4组视网膜取材后进行 NO、NOS生化指标的测定。结果 : ~ 组视网膜组织 NO、NOS含量差异有显著性 ,与 组相比 , 、 组 NO、NOS值显著增高 ;与 组相比 , 、 组 NO、NOS值显著降低 ; 组与 组 NO值统计学差异无显著性意义。 组与 组比较 ,NOS值降低明显。结论 :b FGF有清除自由基、增强抗氧化能力的作用 ,并且高浓度 b FGF较低浓度 b FGF对体外培养的视网膜组织抗氧化作用显著。

【Abstract】 Objective: To study the effect of basic fibroblast growth factor (bFGF) on nitric oxide (NO) and nitric oxide synthase (NOS) in rabbit retina. Methods: Twenty healthy adult rabbits (40 eyes) were randomly divided into 4 groups: Group Ⅰ (fresh retinal tissue), Group Ⅱ (routine culture), Group Ⅲ(high concentrations of bFGF), and Group Ⅳ (low concentrations of bFGF). The retinal tissue in each group other than Group I was cultured for 1 week NO,NOS levels in the retina were detected in all groups. Results: NO,NOS levels in Group Ⅱ were superior to those in other groups. There was no statistical difference in NO content between Group I and Group Ⅲ. NOS levels were significantly decreased in Group Ⅲ, compared with Group Ⅳ. Conclusion: bFGF is able to eliminate free radicals, and enhance the ability of anti-oxidation in retina, especially at high concentration.
