细胞间黏附分子-1在大鼠炎症性肠病模型中的表达研究

【摘要】 目的 :研究细胞间黏附分子 - 1(ICAM - 1)在三硝基苯磺酸 (TNBS) /乙醇所致的大鼠炎症性肠病模型结肠组织中的动态表达情况 ,探讨它在该模型中的作用及意义。方法 :TNBS +乙醇灌肠建立与人类Crohn病相似的急慢性结肠炎的模型 ,分别于 1、3、7、14、2 1d处死 ,通过原位杂交及免疫组化的方法观察结肠组织中ICAM - 1表达情况 ,并比较它与病理改变的关系。设肠道灌注生理盐水组为对照组。结果 :模型组大鼠急慢性期病理改变与人类的Crohn病相似 ,ICAM - 1表达明显增高 ,呈动态改变过程。对照组则很少甚至无表达。结论 :ICAM - 1在TNBS/乙醇模型早期表达开始增多 ,但以慢性期更显著

【Abstract】 Objective: to examine intercellular adhesion molecule 1 (ICAM-1) expression in the colon of 2,4,6-trinitrobenzenesulfonic acid(TNBS)/ethanolinduced model of rats and to explore its effect and significance on pathogenesis of inflammatory bowel disease(IBD). Methods: The rat IBD model were made with TNBS plus anhydrous alcohol and the control group just with normal saline. Then the rats were killed at various time. Hibridization in situ(ISH) and immunohistochemistry(IH) for ICAM-1 were conducted. And the correlation between pathology and hibridization in situ were studied. Results: the inflammation of rats colon is similar to human′s CD. ISH and IH indicate up-regulation of ICAM-1 expression in model group colon with a dynamic curve, but not in the control group. Conclusions: the expression of ICAM-1 is increased in TNBS/ethanol induced colitis, especially in chronic phase.ICAM-1 is very important to pathogenesis of IBD.Anti-ICAM-1 may help to the therapy of IBD.
