大鼠肝脏缺血不同时间再灌注时肝细胞凋亡的动态变化及凋亡相关基因Bcl-2的表达
Dynamic changes of apoptosis of the hepatic cells at different duration of reperfusion after ischemia and the expression of Bcl-2 gene related to apoptosis
【摘要】 目的 :探讨大鼠肝脏经历不同缺血时间后再灌注时肝细胞凋亡、Bcl 2蛋白表达的变化及二者的相关性。方法 :参照Nauta的方法制作大鼠肝脏缺血再灌注损伤模型 ,用流式细胞仪检测肝细胞凋亡百分率及Bcl 2蛋白的表达。结果 :随着肝脏热缺血时间的延长 ,肝细胞凋亡百分率明显增加 ,而Bcl 2蛋白表达量逐渐下降。结论 :肝细胞凋亡在肝脏缺血再灌注损伤过程中发挥着重要作用 ,Bcl 2蛋白对肝细胞凋亡具有抑制作用
【Abstract】 Objective:To explore apoptosis of the hepatic cells and changes of expression of Bcl-2 protein of reperfusion in different duration after hepatic ischemia and the relativity of both.Methods:Model of hepatic ischemia reperfusion injury was established according to Nauta method,apoptosis percentage and the expression of Bcl-2 protein were detected with flow cytometry.Results:The apoptosis percentage of the hepatic cells was increased obviously and the expression amount of Bcl-2 protein was gradually descend with the prolongation of the ischemia duration.Conclusion:Apoptosis of the hepatic cells plays a very important role in the process of the injury of hepatic ischemia reperfusion and Bcl-2 protein can inhibit the apoptosis of the hepatic cells.
