室间隔缺损儿童心房中Ⅰ和Ⅲ型胶原蛋白的表达

【摘要】 目的 探讨室间隔缺损儿童心房Ⅰ型和Ⅲ型胶原蛋白表达的变化。方法 应用SP免疫组化和数字图像分析方法 ,观测 6例正常儿童和 12例室间隔缺损儿童心房Ⅰ型和Ⅲ型胶原蛋白的表达。结果  (1)正常心房Ⅰ型胶原为粗、细不等的条状纤维 ,彼此连接成网 ;Ⅲ型胶原纤维呈散在的斑片状分布 ;(2 )室间隔缺损心房Ⅰ和Ⅲ型胶原表达 ,大多区域属于正常。仅部分区域发生极度重排 ,即Ⅰ型胶原呈现大斑块状增加、断裂或消失 ,Ⅲ型胶原呈条束状增加 ;(3)数字图像分析显示 :正常心房肌Ⅰ型胶原含量较Ⅲ型胶原多 ;室间隔缺损儿童心房肌Ⅰ、Ⅲ型胶原增加显著。结论 室间隔缺损儿童心房部分区域Ⅰ和Ⅲ型胶原纤维重排、含量显著增加。这可能是导致室间隔缺损儿童心功能异常的原因之一。

【Abstract】 Objective To investigate the changes of type Ⅰ and Ⅲ collagen expression in children atrium of ventricular septal defect.Methods To observe and determinate the expression of type Ⅰ and Ⅲ collagen in 6 normal and 12 ventricular septal defect children heart by using immunohistochemistry and digital image analysis technology.Results (1)The type Ⅰ collagen fibers in normal children atrium varied with width and were connected into nets,and type Ⅲ coilagen fibers scattered in speckles.(2)The expressions of type Ⅰ and Ⅲ collagen were mostly normal in children atrium of ventricular septal defect,but a few were redistrbuted severely in some areas.Type Ⅰ collagen fibers increased in large speckles or split or disappeared,and type Ⅲ increased in bundles.(3)The results of digital image analysis showed that type Ⅰ collagen content was much more than that of type Ⅲ in normal children atrium and that the content of type Ⅰ and type Ⅲ collagen increased remarkably in children atrium of ventricular septal defect.Conclusion The expressions of type Ⅰ and Ⅲ collagen were redistribut and increase greatly in some area in children atrium of ventricular septal defect.which may be one of the reasons that caus abnormal heart function in ventricular septal defect.
