米邦塔食用仙人掌试管快速繁殖试验
Culture in Vitro of Edible Cactus Opuntia milpa alta Haw (Cactaceae)
【摘要】 以米邦塔食用仙人掌幼龄茎为外植体 ,研究从诱导培养 ,增殖继代 ,生根培养 ,到瓶外移植各阶段试管快繁的若干影响因子。试验结果表明 ,在附加 3.0mg/LBA +0 .2mg/L NAA的MS培养基上诱导效果较好。 40d后转入附加 2 .0mg/LBA +0 .2mg/L NAA的MS培养基上继代培养 ,以 30d为一个增殖继代周期 ,增殖倍率达 6 .2。随继代次数增加 ,可减少BA用量 ;生根培养阶段以 1 / 2MS +0 .1mg/LNAA的培养基生根效果最好。无机盐浓度减半 ,糖用量减少 1 / 3,有利于生根与壮苗 ;移栽时注意保持水分平衡 ,选择栽植介质 ,控制杂菌污染以及选择适宜的光照和温度条件
【Abstract】 With the young shoots as explants, the effects of different factors on rapid micro propagation from induction, (propagation), rooting to transplant out of vitro of edible cactus Opuntia milpa alta Haw ( Cactaceae) were studied. The results (suggested) that the optimum culture medium for explant of young shoots induction was MS+3.0mg/L BA+0.2mg/L NAA. After 40 days, the adventitious buds were subculturesd in MS+2.0mg/L BA+0.2mg/L NAA medium and the propagation coefficient reached to 6.2 with the 30 days cycles. The BA concentration could be descended when the substructure times added. The medium 1/2MS+0.1mg/L NAA is the best for rooting. Keeping humidity, selecting growing medium, controlling pathogenic bacteria and fungi, taking proper illumination and temperature were important in the stage of transplant out vitro.
