【摘要】 采用改良的CTAB方法提取转基因甘蔗幼苗的基因组DNA ,核酸蛋白测定仪分析及凝胶电泳结果表明 ,提取的DNA具有典型的DNA分子的标准紫外吸收光谱特点 ,DNA产量为 4 5~ 6 0 μg/ 10 0mg。同时根据质粒载体设计合成了 2对引物 ,分别扩增新霉素磷酸转移酶 (nptII)基因和甘蔗花叶病毒外壳蛋白(ScMV CP)基因。PCR检测结果表明 ,在 5 3株待检测的样品中有 2 4株同时含有这 2种转基因成分 ,其中 13株在Southern杂交中有杂交信号出现 ,其拷贝数为 1~ 3个。

【Abstract】 The genome DNA of transgenic sugarcane was extracted by modified method. The ultraviolet absorbance spectrum analysis and the gel electrophoresis results showed that DNA samples possessed the standard ultraviolet absorbance spectrum of pure nature DNA, the yield of DNA obtained by the modified method was 45~60μg/mg young leaves.Two pairs of primers were designed to amplify meomycin phosphotransferase II gene(nptII) and sugarcane mosaic virus coat protein gene(SCMV-CP).The PCR results suggested that only twenty four samples contained two transgenic ingredients among fifty three samples,in which thirteen samples represent southern hybridization signals.And there are one to three copies in each transgenic sugarcane.
