高效液相色谱手性固定相法拆分克伦特罗对映体
Enantioseparation of Clenbuterol Enantiomers by High Performance Liquid Chromatography on Chiral Stationary Phase
【摘要】 以正己烷、异丙醇为流动相 ,用Whelk O1手性固定相成功拆分了药物克伦特罗对映体。出峰时间在 5min左右 ,峰形对称性好 ,完全达到基线分离 ,分离因子大于 1 .4 ,可用于该对映体药物的分析与制备分离。讨论了二元流动相中异丙醇含量及温度的变化对分离的影响 ,并对拆分机理进行了探讨
【Abstract】 The enantiomers of clenbuterol have been originally successfully separated on Whelk O1 chiral stationary phase in hexane/isopropyl alcohol mobile phase with satisfactory results. The two peaks of enantiomers were very symmetric, and the baseline separation was obtained. Furthermore, the retention time about 3~8 min and chiral separation factor α above 1.4 were achieved. The effects of the concentrations of isopropyl alcohol in mobile phase and the temperatures on the chiral separation were investigated experimentally and the mechanism of separation was discussed. The results indicated that the retention time and chiral separation of the analyte varied with the change of mobile phase composition. This method can be used for the analytical and preparative resolution of clenbuterol because of the high value of chiral separation factor α above 1.4.
