新型高效液相色谱酰胺键合固定相的制备与评价
Preparation and Evaluation of A Novel Amide Bonded Stationary Phase for High Performance Liquid Chromatography
【摘要】 将YWG 80硅胶和 3 氨基丙基三甲氧基硅烷反应后与 2 壬基丁二酰氯反应制得一种新型双齿酰胺键合固定相 (BABSP 2 )。采用元素分析和傅里叶变换红外光谱表征了键合相 ;用芳香族化合物溶质和甲醇 水二元流动相 ,考察了键合相的疏水选择性和亲硅醇基活性 ;评估了在酸性条件下 (pH 2 .5 )的水解稳定性。结果表明 :BABSP 2能有效抑制残留硅醇基活性 ,并具有可比的疏水选择性和较好的水解稳定性
【Abstract】 A novel bonded stationary phase, bidenate amide bonded stationary phase (BABSP-2), was prepared by reacting YWG-80 silica gel firstly with 3-aminopropyltrimethoxysilane and then with 2-nonylbutandedioc chloride. Characterization of the prepared packing was carried out with elemental analysis and Fourier transform infrared spectroscopy. Hydrophobic selectivity and silanophilic activity of BABSP-2 were evaluated by using aromatic compounds as analytes and methanol-water as binary mobile phase. Hydrolytic stability of the packing in acidic mobile phase with pH 2.5 was also evaluated. The results show that the novel bidenate amide stationary phase has excellent reversed-phase chromatographic performance and good resistance to hydrolysis. It can be used to separate basic solutes efficiently due to the existence of the bidenate structure containing internal polar amide group contributing to restraining the activity of the residual silanol group.
