白细胞介素-1β对人软骨细胞BaxmRNA表达的作用
The regulating effects of interleukin-1β(IL-1β) on gene expression of Bax mRNA in human hyaline chondrocyte
【摘要】 目的探讨人透明软骨细胞中白细胞介素-1β(IL-1β)对凋亡基因BaxmRNA表达的作用。方法应用反转录-聚合酶链反应(RT-PCR)和荧光定量PCR的方法检测IL-1β作用下传代培养的人透明软骨细胞BaxmRNA的含量。结果对于传代培养人的软骨细胞,IL-1β浓度为1、10、100ng/ml作用12h,对BaxmRNA的调节作用存在剂量依赖关系,各组之间差异有显著性。结论IL-1β可以按照剂量依赖方式正向调节软骨细胞BaxmRNA基因的表达,并通过对细胞凋亡的调控参与骨关节炎的发生、发展。
【Abstract】 Objective To assess the regulating effects of interleukin-1β (IL-1β) on gene expression of Bax mRNA in human hyaline chondrocyte.Methods Hyaline chondrocytes of human were harvested enzymatically and cultured in DMEM supplemented with 20% bovine serum.In the experiment,various concentrations of IL-1β were added to the medium.The effects of IL-1β on the Bax mRNA were assessed by reverse transcriptase polymerase chain reaction (RT-PCR) in the passaged monolayer cell cultures of hyaline chondrocyte.Results IL-1β increased the Bax mRNA level in passaged cultures of hyaline chondrocyte.The difference was significant (P<0.01).The level of Bax mRNA expression was significantly correlated with the dose of IL-1.Conclusion IL-1β can cause a dose-dependent stimulation on Bax gene exression in human chondrocyte.IL-1 may play a crucial role in OA development by regulating chondrocyte apoptosis,indicating that IL-1β may be a target for the gene therapy of OA.
