农杆菌介导获得转水稻草矮病毒NS3基因水稻植株

【摘要】 构建了包含水稻草矮病毒(Ricegrassystuntvirus,RGSV)NS3基因的植物表达载体pCBTNSv3,应用农杆菌介导法,将NS3基因导入水稻愈伤组织中,获得了转RGSVNS3的水稻转基因植株,总DNA经PCR、Southern点杂交鉴定,初步证实NS3基因已整合到水稻基因组中.

【Abstract】 The plant expression vector pCBTNSv3 containing vRNA3 NS3 gene was obtained by replacing the exterior segment of pCBTNSv6 constructed by doctor Zhang Chunmei of our insititute. The recombinant plasmid of pCBTNSv3 was introduced into Agrobacterium tumefaciens by triparental mating. The positive clones were then used in the transformation of rice. By the Agrobacterium-mediated method, the transgenic rice plants were regenerated. PCR and Southern blotting analysis of the total DNA extracted from transgenic plants showed that the NS3 gene of RGSV had been integrated into the rice genome.
