花生幼胚RNA提取方法研究

【摘要】 花生(ArachishypogaeaL.)幼胚体积很小,不易剥取且富含多糖、酚类等化合物,因此以花生幼胚为材料提取RNA时会遇到很大困难.采用改进的Trizol法和异硫氰酸胍一步法成功提取出了高质量的花生幼胚总RNA,后续实验证明具有良好的逆转录活性.介绍了花生幼胚快速剥取的方法.

【Abstract】 It was difficult to isolate total RNA from young peanut embryo, which was not only too small to be obtained, but also contained polysacchrides, phenolic compounds, many unidentified secondary metabolites and so on. Two methods of the extraction of RNA from young peanut embryos, improved Trizol method and one step of guanidiumthiocyanate, were introduced and discussed in this paper. The subsequent experiments proved the high quality of the RNA acquired by the two methods. Meanwhile, the method of rapidly sampling young peanut embryos was also presented.
