重组大豆KUNITZ型胰蛋白酶抑制剂Ti~d的大肠杆菌表达及抑制活性研究
Expression and Activity Study of Soybean Kuntiz-type Trypsin Inhibitor
【摘要】 利用大肠杆菌高效表达系统表达大豆Kunitz型胰蛋白酶抑制剂 (SBTi A2 )Tid,并利用亲和层析的方法纯化得到有抑制胰蛋白酶活性的SBTid.对重组SBTid的 pH稳定性和温度稳定性与天然SBTia进行了比较研究 .结果表明重组SBTid在高 pH下具有更好的稳定性 ,预示了SBTid作为生物农药或利用转基因方法提高作物抗虫性的应用前景
【Abstract】 One kind of Soybean kuntiz-type trypsin inhibitors(SBTi)-Tid was expressed in E.coli and purified by affinity chromatograph. The recombinant Tid showed trypsin inhibitory activity in vitro . The pH stability and thermal stability of Tia and recombinant Tid were studied. Compared with the natural SBTia,SBTid exhibited a better stability in a high pH spectrum and a similar thermal stability at less than 50 ℃ .These data suggested that SBTid has a promising application potential as a bio-pesticide and insect-resistant gene in crop designing.
