原子力显微镜观察Azone复合促进剂对兔舌下粘膜的作用

【摘要】 在接近生理的状态下,用原子力显微镜观察了Azone复合促进剂对离体兔舌下粘膜的作用,以探讨它促进胰岛素经舌下粘膜吸收的机理。结果表明Azone复合促进剂对兔舌下粘膜有显著的作用,而且随着作用时间的延长它对膜的损伤越大。当它与舌下粘膜的作用仅为30min时,就会在粘膜上皮表面形成很多排列整齐的小孔,膜表面的粗糙度(RMS)也显著下降。这些结果说明,Azone复合促进剂一方面通过溶解膜表面的脂类化合物,另一方面通过插入细胞膜的脂质双层中,使膜表面孔径增大,从而改变膜的结构和通透性,促进胰岛素的吸收。

【Abstract】 Atomic force microscopy (AFM) was utilized to examine the influence of Azone composite enhancer (ACE) on rabbit sublingual mucous membrane in vitro. The ACE consists of 5% Propylene glycol, 2.5% Azone and 2.5% Tween 80 in pH 7.4 PBS. The surface image of the sublingual mucous membrane was significantly changed with the exposure time of the membrane to ACE. When the membrane was treated with ACE for 30 min, the surface grain was little out of shape, and a lot of small holes appeared on the membrane. In addition, the surface roughness(RMS)decreased significantly with time. This study indicates that ACE could disrupt the structure of cell membrane by solubilization of lipids, and expanding the size of membrane pore. These changes favor the sublingual absorption of insulin.
