高血黏度对小鼠脑细胞内游离钙浓度及脑组织丙二醛含量的影响
Effects of blood hyperviscosity on cytosolic free calcium concentration in cerebral cells and MDA contents in brain tissue of mice
【摘要】 目的 :探讨慢性脑供血不足对脑细胞内 [Ca2 + ]i,脑组织丙二醛 (MDA)及超氧化物歧化酶 (SOD)含量的影响 .方法 :采用高分子右旋糖酐法建立小鼠高血黏度动物模型 ,以Fluo 3/AM为细胞内游离钙离子的荧光指示剂 ,用激光扫描共聚焦显微镜测定脑细胞内 [Ca2 + ]i 的变化 ;用硫代巴比妥酸法及黄嘌呤氧化酶发光法分别测定脑组织匀浆MDA ,SOD含量 .结果 :高血黏度小鼠脑细胞内 [Ca2 + ]i 明显升高 (P <0 .0 1 ) ,脑组织匀浆中MDA含量明显升高 (P <0 .0 5 ) ,SOD含量明显降低 (P <0 .0 5 ) .相关分析显示小鼠全血高、低切黏度与脑细胞内 [Ca2 + ]i 均呈显著正相关关系 (Spearman等级相关系数分别为 0 .76 9,0 .82 1 ,P <0 .0 1 ) ;全血低切黏度与脑组织MDA含量也呈显著正相关关系 (Pearson相关系数为 0 .72 0 ,P <0 .0 1 ) .结论 :高血黏度可导致脑细胞内钙超载及脑组织中自由基反应增强
【Abstract】 AIM: To investigate the effects of chronic cerebral blood flow shortage caused by blood hyperviscosity on cytosolic free calcium concentration in cerebral cells, MDA and SOD contents in brain tissue. METHODS: Animal model of blood hyperviscosity was established by intravenous injection of high molecular dextran to mice. Alteration of [Ca 2+ ] i in murine cerebral cells was measured by confocal laser scanning microscope using Fluo 3/AM as calcium indicator, and MDA and SOD contents in brain tissue were determined. RESULTS: [Ca 2+ ] i in cerebral cells subjected to blood hyperviscosity increased significantly ( P <0.01) and [Ca 2+ ] i was positively correlated with the level of blood viscosity ( r η H=0.769, r η L=0.821, P <0.01). Compared with those of control, MDA content in the brain tissue rose but the SOD content dropped significantly ( P <0.05). The MDA content in the brain tissue was also positively correlated with the level of blood viscosity ( r η L=0.720, P <0.01). CONCLUSION: Blood hyperviscosity leads to Ca 2+ overload in cerebral cells and the increase of oxygen free radical reaction in brain tissues.
