白细胞介素-5在慢性排斥反应中作用机制的探讨

【摘要】 目的 通过观察白细胞介素 5 (IL 5 )对人嗜酸性粒细胞 (Eos)中转化生长因子TGF β1基因表达的影响 ,以阐明其在Eos相关性慢性排斥反应中的作用。方法 采用改良的密度梯度离心法分离并体外培养人外周血Eos ,以未加任何细胞因子的孔作为阴性对照 ,试验组加入不同浓度的IL 5 ( 10 -1 1 ,10 -9,10 -7mol/L)共培养 ,台盼蓝拒染法观察细胞活力变化 ,培养 16h后收集细胞和细胞培养上清液 ,应用RT PCR和ELISA方法通过检测转化生长因子TGF β1mRNA和蛋白表达的量观察IL 5的调控作用。结果 与对照组 ( 2 2 8.9± 2 .87)相比 ,3种浓度的 ( 10 -1 1 ,10 -9,10 -7mol/L)Th2型细胞因子IL 5均能显著增强体外培养的Eos中TGF β1蛋白的表达 ( 3 3 5 .13± 9.43 ,40 3 .7± 0 .0 9,42 6.0± 0 .0 5 ) ,P <0 .0 5 ,处理组mRNA的量分别为对照组的 1.42 ,1.70和 1.76倍 (P <0 .0 5 )。结论 Th2型细胞因子IL 5可能通过上调TGF β1的基因表达在Eos相关性慢性排斥反应中发挥作用

【Abstract】 Objective To investigate the effects of IL 5 on the expression of TGF β1 in eosinophils in vitro . Methods Eosinophils incubated in vitro in the presence of different concentrations of IL 5 were used as experimentol group and those without IL 5 were used as negative control. The regulatory effect of IL 5 was determined by measuring the expression lovel of TGF β1 RNA in supernatants and cells at 16 h after cultivation using ELISA and RT PCR. Results The expressions of TGF β1 protein (335.13±0.68, 403.7±0.09, 426.0±0.05) in eosinophils were increased significantly by the three different concentrations of IL 5 (10 -11 , 10 -9 , 10 -7 mol/L), while that for control was 228.9±0.92 ( P <0.05). The expression of TGF β1 mRNA in eosinophils was increased significantly by IL 5 in three samples ( P <0.05) as 1.42, 1.70, and 1.76 times as that of the non stimulated controls. Conclusion IL 5 can up regulate the expression of TGF β1 in eosinophils in vitro , which might have effect in eosinophil associated chronic rejection.
