cDNA捕捉法获得水稻杂种不育基因座位Sc区域的编码序列
Isolatation of candidate cDNA clones from rice genomic region covering hybrid sterility locus Sc by cDNA capturation
【摘要】 cDNA捕获法是一种以表达为基础的基因分离技术,直接用目的区域的基因组DNA捕捉该区域编码的cDNA,快速从大的基因组区域分离表达序列.本研究用一个水稻杂种不育基因座位Sc附近的大片段TAC基因组片段来捕捉该区域在水稻穗部表达的cDNA,共获得了6条不同的cDNA.将这些cDNA克隆进行测序分析,获得了该区域在水稻穗部表达的部分基因,其中1个是籼稻特有的基因.这些cDNA片段可成为新的标记用于目的基因的精细定位和候选基因序列分析.
【Abstract】 cDNA capturation is an expression-based gene isolation technique that uses genomic DNA to capture cDNAs directly and thus can rapidly identifies expressed sequences from large genomic region.By means of this technique a captured cDNA library of rice immature spikelet was constructed using two TAC clones M90J01 and M45E14 as the anchor DNAs,which cover a hybrid sterility locus Sc on chromosome 3.More than 20 positive clones were obtained from the captured cDNA libraries.By sequencing and database search analysis,the cDNAs were identified as six different genes,of which one is indica-specific gene.These genes can be used as new markers for fine-mapping of Sc,as well as candidates of the Sc gene.The results indicated that this method is useful for positional cloning and constructing detailed transcription maps of target gene regions.
