干细胞因子对大鼠骨髓间充质干细胞增殖与分化的影响

【摘要】 目的 探讨干细胞因子 (StemCellFactor ,SCF)促进骨髓间充质干细胞 (MSCs)增殖及促进其向心肌细胞分化的作用。方法 SCF对MSCs预处理 ,流式细胞术测定第 4代MSCs的细胞周期 ,DAPI -MSCs在体外模拟的心肌微环境下培养。用数码显微摄像及免疫荧光技术分别记录和检测心肌特异性肌节肌球蛋白重链 (MHCα/β)、肌钙蛋白T(TroponinT ,TnT)的表达 ,分析MSCs向心肌细胞分化的百分率。结果 实验组的G0 /G1期细胞百分率均显著低于对照组 (P <0 0 1) ,S期、G2 /M期细胞百分率及PI值均显著高于对照组 (P <0 0 1)。在共培养的第 2、3、4、5d ,SCF组表达MHC的DAPI -MSCs阳性百分率分别为 (1 96± 0 3 3 ) %、(4 76±0 3 2 ) %、(5 15± 0 42 ) %、(8 5 2± 0 45 ) % ,显著高于对照组的 (0 5 2± 0 2 1) %、(1 2 5± 0 47) %、(1 75± 0 2 8) %、(3 64± 0 3 2 ) % (P<0 0 1) ;在共培养的第 3、4、5d表达TnT的DAPI -MSCs阳性百分率分别为 (1 10± 0 15 ) %、(2 64± 0 18) %、(5 43± 0 2 4) %均显著高于对照组的 0、(1 2 7± 0 13 ) %、(1 3 2± 0 2 5 ) % (P <0 0 1)。结论 SCF对MSCs具有促增殖、促分化的作用。

【Abstract】 Objective To investigate the effect of stem cell factor (SCF) on the proliferation and differentiation of mesenchymal stem cells (MSCs) into cardiomyogenic cells in rats. Methods The MSCs pretreated by SCF were cocultured with cardiomyocytes. The cell cycle of 4th passage MSCs was analyzed by flow cytometry. The morphologic changes of MSCs were recorded with digital microscope camera system, and the expressions of cardiac myosin heavy chain (MHCα/β) and troponin T (TnT) were detected by immunofluorescence technique. The percentage of the differentiation of MSCs into cardiomyogenic cells was calculated. Results The percentage of G 0/G 1 phase cells of MSCs in the SCF group was significantly lower than that in the control group(P<0.01). The percentage of S and G 2/M phase cells as well as the proliferation index of MSCs in the SCF group was significantly higher than that in the control group(P<0.01).The percentages of MHC protein-positive MSCs in the SCF group[(1.96±0.33)%,(4.76±0.32)%,(5.15±0.42)%,(8.52±0.45)%] were significantly higher than those in the control group[(0.52±0.21)%,(1.25±0.47)%,(1.75±0.28)%,(3.64±0.32)%] at the 2th,3th,4th,and 5th days, respectively(P<0.01). The percentages of TnT protein-positive MSCs in the SCF group [(1.10±0.15)%,(2.64±0.18)%,(5.43±0.24)%]were significantly higher than those in the control group[0,(1.27±0.13)%,(1.32±0.25)%]at the 3th,4th, and 5th days, respectively(P<0.01). Conclusion SCF could stimulate the proliferation of MSCs and induce MSCs to differentiate into cardiomyogenic cells in vitro.
