【摘要】 目的 评价阿托伐他汀 (atorvastatin)对脂多糖 (LPS)诱导下人肺上皮细胞环氧化酶 -2 (COX -2 )表达及蛋白分泌的影响。方法 对LPS诱导的人肺上皮细胞株 (A5 49)给予不同浓度的阿托伐他汀干预 ,同时设置对照组 ,培养 12h后用Tris法处理细胞后提取RNA用于RT -PCR(reversetranscriptase -polymerasechainreaction)及留取细胞用于WesternBlot分析。结果阿托伐他汀明显降低LPS诱导下COX -2mRNA的表达及COX -2蛋白的分泌 (P <0 0 5 ) ,且表现浓度梯度的变化。结论 阿托伐他汀降低降低脂多糖 (LPS)诱导下人肺上皮细胞环氧化酶 -2 (COX -2 )的表达

【Abstract】 Objective To assess the effect of atorvastatin on the Lipopolysaccharide (LPS)-induced cyclooxygenase-2 (COX-2) expression in cultured human pulmonary epithelial cells (A549). Methods A549 cells were incubated in the medium containing LPS and different concentrations of atorvastatin(0,10,15,20μM/ml, respectively) for 12h. Then the total cellular RNA and proteins from the cells treated with different experimental conditions were extracted for RT-PCR and western blot analysis,respectively. Results In cultured human pulmonary epithelial cells, atorvastatin reduced the expression of COX-2 mRNA and protein induced by LPS in a dose-dependent manner. Conclusion Atorvastatin may down-regulate LPS-induced COX-2 expression in cultured human pulmonary epithelial cells.
