家蚕丝素固定化链霉菌的生物化学特性

【摘要】 脱胶蚕丝经不同处理制得多孔碱化丝素和丝素粉末 ,采用吸附和戊二醛交联法将链霉菌 (StreptomycesflavovirensA8)固定 ,制得不同的固定化链霉菌 (Ⅰ、Ⅱ、Ⅲ )。实验结果表明 :其固定化链霉菌Ⅰ、Ⅱ和Ⅲ的米氏常数分别为Km1=2 4 0 73× 10 -2 mol/L ,Km2 =2 5 36× 10 -2 mol/L ,Km3 =2 772 3× 10 -2 mol/L ,而游离链霉菌的Kmf 为2 3715× 10 -2 mol/L ;固定化链霉菌Ⅰ的酶活力半衰期在 6 4d以上 ,固定化链霉菌Ⅱ和Ⅲ的半衰期最长达 6 8d ;固定化链霉菌对尿素有较强的抵抗能力 ;Co2 + 、Mg2 + 对链霉菌葡萄糖异构酶有激活作用 ,而Fe3 + 和Ca2 + 是葡萄糖异构酶的抑制剂。

【Abstract】 Basified fibroin and powdery fibroin was prepared by different treatment methods using degumed silk.The Streptomyces flavovirens A8 was immobilized on the fibroins by using adsorbing and cross-linking agent to form the immobilized S.flavovirens A8 Ⅰ,Ⅱand Ⅲ.The experiment results showed that these immobilized Streptomyces flavovirens A8s’ Michaclis constant were Km 1=2.407 3×10 -2 mol/L,Km 2=2.536×10 -2 mol/L,Km 3=2.772 3×10 -2 mol/L respectively,and the free Streptomyces flavovirens A8s’ Michaclis constant was Km f=2.371 5×10 -2 mol/L.The result also indicated that the work half-life of the immobilized Streptomyces flavovirens A8 Ⅰ was 64 days and the Ⅱ and Ⅲ were 68 days.The immobilized Streptomyces flavovirens A8s had more strong ability to resist urea solution.Experiment also showed that Co 2+ ion and Mg 2+ ion were activators and Ca 2+ ion and Fe 3+ ion were inhibitor to immobilized Streptomyces flavovirens A8s.
