家蚕脂肪体组织基因表达谱的研究 Ⅰ.家蚕5龄中期幼虫脂肪体组织基因表达分析

【摘要】 以大规模EST测序为基础 ,用生物信息学方法分析了家蚕 5龄 3d脂肪体组织基因表达的特征 ,共获取脂肪体组织EST 12 72 1个。经Phrap程序拼接得到 2 316个非冗余序列 ,其中包括 82 4个contigs,14 92个singletons。在鉴定出的已知基因中 ,基因表达频率有明显差异 :物质代谢相关基因表达量最高 ,占总量的 5 8 2 % ;其次为转录翻译调节相关基因和酶类基因 ,分别占 15 5 %和 14 5 %。在高量表达基因中 ,有贮藏蛋白、蛋白酶抑制剂、类IDGF蛋白、抗菌蛋白、谷胱甘肽 S转移酶等。结果表明 ,脂肪体组织基因表达的种类很多 ,特征明显 ,与脂肪体在蚕体内的贮藏功能、蛋白质合成功能、代谢调节功能以及变态发育有密切关系。

【Abstract】 The gene expression profile of the fat body of the 3 rd day of the 5 th instar larvae of Bombyx mori (Dazao) was analyzed with a combination of massive EST sequencing and bioinformatics. All together, 12 721 ESTs in the fat body were obtained. After processing of the ESTs with software Pharp, 2 316 unigenes were identified. Among them 824 were contigs and 1 492, singletons. The expression frequencies of the identified known genes were clearly in great disparities. The expression level of the substance metabolism-related genes was the highest, accounting for 58.2% of the total; the expression level of the transcription-and translation-related and enzyme genes accounts for 15.5% and 14.5% respectively. Among those abundantly expressed were storage proteins, protease inhibitors, IDGF like proteins, antibacterial peptides, glutathione-S transferase, etc. These results show that in fat-body many genes were expressed, with distinct features. This expression profile is closely related to its functions in storage, protein synthesis, metabolic regulation and metamorphosis.
