飞天蜈蚣七对实验性肝纤维化大鼠胰岛β细胞功能的影响

【摘要】 目的 探讨应用飞天蜈蚣七抗肝纤维化时对大鼠胰岛β细胞功能的影响。方法 以四氯化碳诱导大鼠肝纤维化模型,设立正常组,模型组和飞天蜈蚣七组。正常组于实验开始时取胰腺,模型组和飞天蜈蚣七组则分别于造模后3周,6周取胰腺作胰岛细胞的分离与培养,然后测定其培养上清液胰岛素含量。结果 3周模型组大鼠空腹血清胰岛素与正常组差异无显著意义,而6周模型组大鼠空腹血清胰岛素为(53.8±3.1)mU/1,正常组为(24.5±2.5)mU/1,两组比较q=12.87,P<0.05;3周模型组大鼠胰岛细胞的基础胰岛素分泌与正常组差异无显著意义,而6周模型组大鼠胰岛细胞的基础胰岛素分泌量为(50.90±12.8 6)μU/ml,正常组为(34.87±5.16)μU/ml,两组比较q=11.08,P<0.01;3、6周飞天蜈蚣七组大鼠胰岛细胞的基础胰岛素分泌量分别为(156.01±7.11)μU/ml和(141.43±39.01)μU/ml,均高于相应模型组,F值分别为65.80和12.66,P<0.01。结论 飞天蜈蚣七可增加四氯化碳所致肝纤维化3、6周大鼠胰岛β细胞的基础胰岛素分泌量。

【Abstract】 Objective To investigate the effect of Aralia chinesis. L. on pancreatic islet B cells of experimental hepatic fibrogenesis rats. Methods Rats were randomly allocated into three groups; normal group, model group and Aralia chinesis. L. group. The rats in the latter two groups were administered with CCl4 solution to induce liver fibrosis, the Aralia chinesis. L. group was also treated with Aralia chinesis. L. 10 days after the beginning of CCl4 administration, Rats in normal group were sacrificed at the beginning of the experiment, while the rats in the other two groups were sacrificed randomly at the end of the third and sixth weeks. The rats’ islets were isolated and cultured in vitro, then the basal insulin level of the islets and the serum level of the islets and the serum level of insulin were determined by radioimmunological assay. Results It seemed no change that the levels of serum insulin and basal insulin between the model group and the normal group at the third week. But at the sixth week, both insulin levels in the model group were higher than those in the normal group (53.8 ± 3. 1mU/L VS 24.5 ± 2. 5mU/L, Q = 12.87, P<0.05; 50.90±12.86μU/ml VS 34.87±5.16μU/ml, Q = 11.08, P<0.01). But the basal insulin levels in the Aralia chinesis.L. group were apparently higher than those in the model group at the third and sixth weeks (156.01 ± 7. 11μU/ml VS 39.84 ±4.01μU/ml, Q=65.80, P<0.001; 141.43±39.01μU/ml VS 50.90±12.86μU/ml, Q = 12. 66, P<0.01). Conclusion Aralia chinesis. L. can increase the basal insulin level of the islets in CCL4-induced liver fibrosis rats.
