COL1A1基因-1997G/T多态性与中国女性骨密度关系分析
The Relationship Between -1997G/T Polymorphism of COL1A1 Gene and Bone Mineral Density in Chinese Women
【摘要】 目的 :研究COL1A1基因启动子区 -1997G/T单核苷酸多态性与中国女性骨密度关系。方法 :采用双能X线吸收仪对 2 91名女性研究对象进行腰椎及股骨扫描 ,应用PCR -限制性片段长度多态性 (PCR -RFLP)方法检测COL1A1基因 -1997G/T多态位点基因型 ,用广义线性模型分析与腰椎 2~ 4及股骨骨密度关系。结果 :在调整环境危险因素前后 ,均未发现COL1A1基因的 -1997G/T位点的多态性与女性股骨颈、股骨柄和股骨三角区及腰椎 2~ 4的骨密度呈显著相关。结论 :在中国女性人群中COL1A1基因 -1997G/T多态性与股骨及腰椎骨密度无显著性相关 ,与欧洲人群研究结果不同。
【Abstract】 Objective:In order to investigate the association of–1997G/T polymorphism in the upstream regulatory region of collagen type I alpha 1 (COL1A1) and Bone Mineral Density (BMD) in Chinese women. Methods:The bone mineral density of lumbar spine (L2~4) and femur bone was measured by dual-energy X-ray absorptiometry in 291 study subjects. PCR-restriction fragment length polymorphism (PCR-RFLP) was applied to examine COL1A1–1997G/T polymorphism of the COL1A1 gene. Generalized linear model was used to analysis the association of this polymorphism and the bone mineral density of the lumbar spine (L2~4) and femur. Results:Without or with adjustment for environmental covariates, no significant correlation was found between the polymorphism -1997G/T of the COL1A1 gene and BMD, including Femoral neck BMD, Ward triangle BMD and Femoral shaft BMD. Conclusion:This results suggest that there is no significant relationship between COL1A1 -1997G/T Polymorphism and femoral BMD and lumbar spine BMD in Chinese women population. Our result is different from that in European population.
