川芎嗪和剪切率对大鼠悬浮微血管内皮细胞凋亡和功能的影响

【摘要】 以川芎嗪和锥 板旋转剪切方式共同作用于悬浮状态的大鼠脑微血管内皮细胞 (rCMEC) ,采用AnnexinV FITC PI双标记流式细胞仪检测细胞凋亡率和测定剪切液中t PA、PAI、 6 K PGF1α、TXB2 、vWF以探讨川芎嗪和剪切率对悬浮血管内皮细胞凋亡和功能的影响。结果表明 :川芎嗪 (31 5、 6 3 0、 1 2 6 0 μg ml)和剪切率(2 0 0 0、 4 0 0 0、 6 0 0 0、 80 0 0s- 1 )对悬浮rCMEC的凋亡和功能有显著影响 ,在对各指标的影响上 ,二者均无交互作用 ;随着剪切率的逐渐增加 ,rCMEC的生存状态和生理功能渐趋紊乱 ,表现为凋亡率增加 ,t PA和PGI2 分泌量减少 ,PAI、TXB2 和vWF分泌量增加 ;给予川芎嗪后可抑制高剪切率诱导的rCMEC凋亡 ,促进rCMEC分泌t PA和PGI2 ,抑制PAI、TXB2 和vWF的分泌。结果提示 :高剪切率可诱导rCMEC凋亡 ,导致悬浮rCMEC功能紊乱。川芎嗪可明显抑制其凋亡和功能紊乱

【Abstract】 Objective The study aimed at the effects of tetramethylparazine (TMP) and shear rate on the apoptosis and functions of microvascular endothelial cell in suspension. Methods Cultured rat cerebral microvascular endothelial cell (rCMEC) was investigated by administration of TMP incorporated with different levels of shear rate introduced by a rotational rheometer. The apoptosis was determined by Annexin V/PI with flow cytometer. The levels of t PA and PAI, 6 K PGF 1α, TXB 2 and vWF in the medium were measured accordingly. Results Both TMP(31^5、63 0、126 0μg/ml) and shear rate (2000、4000、6000、8000s -1) influenced the apoptosis and functions of suspending rCMEC significantly. No interaction for the two factors was found. With elevation of shear rate,the apoptosis rate of rCMEC gradually increased, the function of suspending rCMEC was gradually disordered, shown by the reducing secretions of t PA and PGI-2, and elevating secretions of PAI,TXB 2 and vWF. TMP of 31 5,63 0,126 0μg /ml could improve secretions of t PA and PGI 2 of the suspending rCMEC, but reduce the secretions of PAI TXB-2 and vWF. Conclusion TMP may reduce apoptosis of suspending rCMEC induced by high shear rate and protect the functions of the cells.
