【摘要】 本文建立了一种经改进能特异的、敏感的测定人Ⅲ型前胶原 (hPCⅢ )含量的RIA法 ,并用于诊断肝硬化的临床评价。从人工流产胎皮中分离、纯化hPCⅢ ,免疫家兔获取高效价抗血清。hPCⅢ标记1 2 5I,使用双抗体+PEG分离技术 ,平衡法加样 ,建立RIA。结果表明 ,标准曲线测定范围为 4 0 - 6 4 0 μg/L。与hPCⅠ的交叉反应率为 0 .81 %、与CⅣ、CⅢ、CⅠ无交叉反应。灵敏度为 6 μg/L。精确度 :批内CV为 4 .2 % ;批间CV为 8.2 %。平均回收率为 99.9%。血清hPCⅢ正常参考值为 87.5± 1 6 .2 μg/L ,正常上限值为 1 2 0 μg/L。本文建立的改良法操作更简便、快捷。更多还原

【Abstract】 A specific and sensitive RIA for the human procollagen type Ⅲ (hPC Ⅲ)was developed by double antibody and PEG technique which allowed easy and sequential measurement of hPC Ⅲ in the serum of human.The hPC Ⅲ was isolated and purified from human fetal skin of induced abortion. The antiserum with high titre was acquired from rabbit after immunization.hPCⅢ was labeled with 125 I. The standard curve had good competitive inhibition, and its working range was 40-640μg/L.There was no cross-reaction with hPCⅠ and collagen type Ⅳ or type Ⅲ or type Ⅰ.The intrassay CV was 4.2% and the interassay CV was 8.2%.The sensitivity was 6μg/L and the recovery was 97.5%-106.3%(Mean 99.96%).The normal value of reference was 87.5±16.2μg/L and the upper limit was 120μg/L.The operation of improved RIA for hPCⅢ was simpler and faster than old one.更多还原
