牛磺酸和支链氨基酸对大鼠肝细胞氧化损伤的保护作用

【摘要】 ①目的 探讨牛磺酸和支链氨基酸对CCl4致大鼠体外肝细胞损伤的保护作用及其可能机制。②方法 实验分为对照组、损伤组、牛磺酸 (Tau)组、牛磺酸和支链氨基酸 (T&B)组。用 1 0、2 0mmol/LCCl4诱导大鼠体外肝细胞过氧化损伤 ,并以 1 .5 0mmol/LTau和按一定比例混合的T&B复合物 0 .75 g/L进行肝细胞保护。测定孵育液中谷丙转氨酶 (ALT)、谷草转氨酶 (AST)活性及丙二醛 (MDA)含量。③结果 以 1 0mmol/LCCl4诱导肝细胞损伤时 ,Tau和T&B均能降低血清ALT、AST活性 ,减少MDA生成 ,与损伤组相比有显著差异 (F =4 .4 4 5~1 4 .0 71 ,q =3.82 5~ 4 .6 72 ,P <0 .0 1 )。以 2 0mmol/LCCl4诱导肝细胞损伤时 ,T&B能降低血清ALT、AST活性 ,减少MDA生成 ,与损伤组相比有显著差异 (q =4 .933~ 6 .0 0 1 ,P <0 .0 1 ) ,而Tau只能降低ALT、AST活性 (q =5 .6 0 3、5 .986 ,P <0 .0 1 ) ,但对MDA的生成影响不大 (q =1 .879,P >0 .0 5 )。 ④结论 牛磺酸和支链氨基酸对CCl4所致的肝细胞损伤具有保护功能 ,其保护作用可能是通过抗脂质过氧化作用实现的。

【Abstract】 Objective To study the protective effect of taurine and branched chain amino acid(T&B) on the lipid peroxidation in cultured hepatocellular injury. Methods The experiment was performed in four groups: control; injury; taurine and T&B groups. Lipid peroxidation in cultured hepatocellular injury was induced by 10 mmol/L and 20 mmol/L carbon tetrachloride(CCl 4). T&B was mixed in proper proportion and used to protect the cultured hepatocytes. ALT, AST and MDA in the cultured liquid were determined to show the hepatocellular function and the degree of lipid peroxidation. Results When the injury was induced by 10mmol/L CCl 4, both taurine and T&B could reduce the activity of transaminase and the content of MDA ( F= 4.445-14.07,q=3.825-4.672,P<0.01). When induced by 20 mmol/L CCl 4 ,significant difference was noticed in the activity of ALT, AST and the content of MDA between the T&B group and the injury group (q=4.933-6.001,P<0.01), while taurine could only reduce the activity of transaminase but had no effect on MDA content (q=5.603, 5.986,P<0.01,q=1.879, P> 0.05). Conclusion Taurine and branched chain amino acid can protect the hepatocytes injured by CCl 4. The possible mechanism is the prevention of lipid peroxidation in cultured hepatocellular injury.
