不同营养液液体培养对交织顶孢霉形态和胞外多糖的影响

【摘要】 通过改变蔗糖浓度和选用不同的氮源对交织顶孢霉RCEF0260菌株液体发酵工艺进行研究。结果表明,不同的氮源对菌丝体的形态影响显著。以蛋白胨和黄豆粉为氮源时,菌丝较粗,分生孢子较宽,呈卵形、长卵形至椭圆形,且黄豆粉组可见大量分生孢子和具长芽管的孢子;而以硝酸铵为氮源时,菌丝细长,孢子一般较窄,呈梭形。不同浓度的蔗糖对菌丝体的形态影响不大,但对胞外多糖产量的影响显著。另外,不同氮源对菌重的影响显著。

【Abstract】 In the paper, the liquid culture of Acremonium implicatum was studied by means of different sucrose concentrations and nitrogen sources. The results showed that different nitrogen sources all had obvious effects on the morphologies of the mycelia. When peptone and soybean powder were of nitrogen sources, the diameter of mycelium was thicker and the conidia, in shape of oval, long oval or oblong, wider than that of NH4NO3, under which having fusiform conidia. Meanwhile, many conidia, some of them with long germ tubes, could be observed when soybean powder was used as nitrogen source. The effects of different concentrations of sucrose on the mycelial morphology was not obvious, but the exo-polysaccharides yield was significant. In addition, the effect of different nitrogen sources on the mycelial biomass was significant.
