红酵母细胞中β-胡萝卜素的快速分离柱高效液相色谱法测定

【摘要】 研究了用快速分离柱高效液相色谱法测定红酵母细胞中β-胡萝卜素的方法。发酵法得到的样品经匀浆后用四氢呋喃提取,提取液以ZORBAXStableBound(4.6×50mm,1.8μm)C18快速分离柱为固定相,乙腈-四氢呋喃体积比8020为流动相分离,流速为1.5ml/min;在该色谱条件下,β-胡萝卜素在1.0min内可达到基线分离;用紫外二极管矩阵检测器检测。方法标准回收率为98%~103%,相对标准偏差为0.46%~0.58%,可用于几种发酵样品中β-胡萝卜素的测定,结果可靠。

【Abstract】 A rapid column high performance liquid chromatography (HPLC) method for determination of β-carotene in red yeast (Rhodotorula glutinis) was studied. The β-carotene was extracted from the sample with tetrahydrofuran by ultrasonic extraction. Then the β-carotene was separated on a ZORBAX Stable Bound (4.6×50 mm, 1.8μm) rapid chromatographic column with acetonitrile-tetrahydrofuran (80:20) as mobile phase at a flow-rate of 1.5 ml·min -1 , and monitored with the photodiode array detector at 451 nm. The recoveries of the β-carotene were 98% to 103%, and the relative standard deviations were 0.46% to 0.58%. This method was applied to the determination of β-carotene in red yeast with good results.
