侵染西瓜的西瓜花叶病毒生物学鉴定

【摘要】 用生物学、血清学及电镜等技术鉴定了侵染西瓜花叶病毒宿州分离物的基本属性。研究结果证明 ,该分离物侵染西瓜、南瓜等葫芦科植物并表现花叶 ;在苋色藜、千日红表现局部褪绿斑 ;桃蚜可传毒 ,该分离物汁液的致死温度为 5 5~ 5 8℃ ,稀释限点为10 - 3~ 10 - 4,体外保毒期为 6~ 10d。病毒粒体为线状 ( 73 0nm× 16nm)。该分离物的提纯物紫外光最高吸收峰在 2 60nm处 ,最小在2 40nm处。A2 6 0 /A2 80 =1.3 2。其病毒的RNA含量约为 8%。ELISA测定该分离物与西瓜花叶病毒 2号 (WMV -2 )的抗血清学反应为阳性。初步确定该分离物隶属WMV -2的一个株系。

【Abstract】 s In this paper, the basic properties of the isolation substance from WMV-2 were identified with the methods of biology, serums and electric microscope. The results showed that the isolation substance could infect Cucurbita modchate, Cucurbitapeno which belonged to calabash subject, and so on, and displayed mosaic. In addition, other plants such as Chenopodium anarenticolor and Gompgrema globosa showed green-fading spots. On the other hand,Myzus persicae and Aphis gossypii could spread poison to other plants. Thermal inactivation point(TIP) of the isolation substance liquids was 55~58*!℃.Dilution end point(DEP) was 10 -3 ~10 -4 and longevity in vitro(LIV) was 6~10*!days. The shape of the virus was linear and its length width=730*!nm×16*!nm,A 260 /A 280 =1.32. RNA content was about 8%. The serum reaction between the isolation substance and WMV-2 was positive by ELISA. It was primarily confirmed that the isolation substance belonged to WMV-2.
