农杆菌介导AFP1基因转化小麦获得转基因植株
AFP1 gene transformation to wheat by Agrobacterium tumefaciens
【摘要】 以nptⅡ基因为筛选基因,利用农杆菌转化系统向普通小麦品种核生3号和99P的愈伤组织中转入白粉病抗性基因Rs-AFP1;获得抗生素抗性克隆数目分别为27个和24个,其中再生绿苗的克隆分别为2个和8个;PCR分析检测所得nptⅡ阳性植株分别为2株和6株,AFP1基因阳性植株分别为1株和4株。
【Abstract】 Powdery mildew resistant gene AFP1 was transformed into the calli of wheat (Triticum aestivum L.) cultivar Hesheng3 and 99P by Agrobacterium tumefaciens), and nptⅡ gene was used as selective marker. After cultured on the selective media, paromomycin resistant calli were got at the number of 27 and 24 respectively; 3 and 8 green shoots were regenerated from the paromomycin resistant calli respectively; PCR analysis proved that the (nptⅡ) positive plants were 2 and 6 respectively, and AFP1 positive plants were 1 and 4 respectively.
