【摘要】 目的:观察8种中草药(雷公藤、白花蛇舌草、黄柏、黄芩、茯苓、红花、板蓝根、丹参)煎剂对银屑病角质形成细胞内 C-erbB-1癌基因表达的抑制作用,探讨中草药治疗银屑病可能的作用机理。方法:8种中草药分别与体外培养的银屑病角质形成细胞和经银屑病患者外周血白细胞刺激的正常人角质形成细胞共同培养,采用细胞原位杂交的方法检测细胞中 C-erbB-1mRNA 的表达。结果:8种中草药对银屑病患者角质形成细胞内 C-erbB-1的表达有不同程度的抑制作用(P<0.001),抑制作用最强的为雷公藤;中药对经银屑病患者外周血白细胞刺激的正常人角质形成细胞内 C-erbB-1的表达也有不同程度的抑制作用(P<0.001),抑制作用最强的为茯苓。结论:这些中药可能通过抑制银屑病角质形成细胞 C-erbB-1的表达来抑制表皮细胞的增殖。更多还原

【Abstract】 Objective:To study the effect and mechanism of 8 kinds of Chinese herbal decoctions in treating psoriasis with observation of in- hibitory effect on expression of C-erbB-1 in psoriatic keratinocytes.Methods:8 Chinese herbal decoctions (Tripterygium wilfordii, Oldenlandia diffusa,Phellodendron chinense,Astragalus membranaceus,Poria cocos,Carthamus tinctorius,Isatidis indigotica,Salvia miltiorrhizae) were observed,which were separately cultured with psoriatic keratinocytes and normal keratinocytes stimulated with psoriatic leucocytes from peripheral blood.The expressions of C-erhB-1 mRNA in the cultured keratinocytes were examined in situ hybridization.Results:The 8 kinds of herbal decoctions could inhibit the expressions of C-erbB-1 in psoriatic keratinocytes in various degree (P<0.001),and the inhibitory effect of Tripterygium wilfordii is the most potent than that of other herbs.The 8 kinds of herbal decoctions could also inhibit the expression of C-erbB-1 in normal keratinocytes stimulated with psoriatic leukocytes (P<0. 001) and the inhibitory effect of Poria cocos is the strongest than that of others.Conclusion:These herbal decoctions could inhibit the proliferation of keratinocytes,which is possibly through the effect on the expression of C-erbB-1.更多还原
