补肾益气活血方药对胎儿宫内生长迟缓胎鼠脑葡萄糖转运蛋白基因表达的影响
Effect of Bushen Yiqi Huoxue Recipe on Gene Expression of Glucose Transporter Protein in Fetal Brain of Intrauterine Growth Retardation Rats
【摘要】 目的:探讨补肾益气活血方药防治胎儿宫内生长迟缓(IUGR)的分子机制。方法:以被动吸烟法建立 IUGR 的动物模型,分为正常对照组(正常组)、IUGR 组、中药治疗组(中药组)、及左旋精氨酸治疗组(精氨酸组)。检测了各组胎鼠脑的重量、胎鼠血清葡萄糖水平以及胎鼠脑中葡萄糖转运蛋白(GLUT)家族中 GLUT1、GLUT3 mRNA 水平和蛋白浓度,其中 mRNA 水平和蛋白浓度分别利用逆转录-聚合酶链反应(RT-PCR)和 Western blot 的方法进行检测。结果:IUGR 组和精氨酸组脑的重量、胎鼠血清葡萄糠水平较中药组和正常组明显降低(P<0.05),而中药组和正常组之间无明显差异。胎脑 GLUT1 mRNA 水平在 IU-GR 组中明显升高,其余三组间无明显差异;而 GLUT3 mRNA 水平在中药组中明显高于 IUGR 组(0.9458±0.1448 vs 0.7951±0.1534 P=0.0365)。GLUT3和 GLUT1(55KD)的蛋白浓度在 IUGR 组及精氨酸组中都明显降低,中药组和正常组之间差异无显著性。GLUT1(45KD)的蛋白浓度在4组之间均无差异。结论:胎鼠血清葡萄糖水平降低及 GLUT 蛋白在胎鼠脑中的低表达在 IUGR 发生中有重要作用;补肾益气活血方药通过提高 IUGR 胎脑 GLUT3 mRNA 对低能量供给的反应,并通过在转录后水平增强 GLUT1和 GLUT3基因的表达,加强胎脑对葡萄糖的摄取利用,为其提供足够能量从而促进脑的发育,达到防治 IUGR 的目的。
【Abstract】 Objective:To explore the molecular mechanism on using Bushen Yiqi Huoxue Recipe (BYHR) in treating intrauterine growth retardation (IUGR).Methods:The rats IUGR model was established by passive smoking.Forty cases were divided into 4 groups: the normal control group,BYHR treated group,IUGR model group and L-Arginine (L-Arg) treated group.The glucose transporter Ⅰ (GLUT1) and glucose transporter 3 (GLUT3) mRNA and protein level were examined in fetal brain by reverse transcriptase poly- merase chain reaction (RT-PCR) and Western blot (WB) respectively.The fetal brain weight and serum glucose concentration were also measured.Results:The fetal brain weight and serum glucose concentration in IUGR group was significantly lower than those of control group (0.144±0.014g vs 0.167±0.013g,P<0.05;2.74±0.87mmol/L vs 5.29±1.53mmol/L,P<0.01;insignifi- cant difference was found between the BYHR and control group.The increase of GLUT1mRNA was detected in IUGR group,but the concentration of GLUT3mRNA in BYHR group was much higher than that of IUGR group (0.9548±0.1448 vs 0.7951±0. 1534,P=0.0365).The protein concentration of GLUT1 (55KD) and GLUT3 decreased significantly in IUGR group and L-Arg group,while there was insignificant difference of the GLUT1 (45KD) levels among the four groups.Conclusion:The low concentra- tion of serum glucose and GLUT protein in fetal brain might play an important role in occurrence and development of IUGR.The possible molecular mechanism in treating IUGR with BYHR might be through erhancing the response of the GULT3mRNA in fetal brain to low energy supply at critical period of brain maturation,but also through enhancing expression of GLUT1 and GLUT3 gene at the post-transcriptional level.
