【摘要】 目的:在临床研究和动物实验的基础上,进一步探讨健骨二仙丸对成骨细胞分化的影响。方法:分离、培养人的原代成骨细胞,采用 PNPP 法、RIA 法、茜素红染色法以及定量 RT-PCR 之法分别检测碱性磷酸酶活性、骨钙素含量、矿化结节形成和Ⅰ型前胶原 mRNA 的表达,从而了解健骨二仙丸对成骨细胞分化的影响。结果:健骨二仙丸中、高剂量能显著增加碱性磷酸酶的活性及矿化结节形成的数量,促进成骨细胞骨钙素和Ⅰ型前胶原 mRNA 的表达(P<0.01,P<0.05),与雌激素(倍美力)组比较差异无显著性。结论:健骨二仙丸能有效促进成骨细胞的分化。更多还原

【Abstract】 Objective:Based on the clinical research and animal experiment results,we intend to explore the effect of Jiangu Er’ xian Pill (JEP) on the differentiation of osteoblast-like cells.Methods:Human osteoblast-like cells were employed,and they were isolated and cultured according to the references.The differentiation of human osteoblast-like cells was observed by the activity of alkaline phos phatase (ALP) and osteocalcin,the number of mineral nodules,and expressing of procollogen 1 α mRNA.The method of PNPP, radioimmunoassay,alizarin staining and quantity RT-PCR were used respectively.Results:The activity of ALP and osteocalcin of JEP groups of the moderate and high dosage are significantly higher than those of control groups.The numbers of mineral nodules, and the expression of procollagen Ⅰα mRNA of the two groups are also significantly enhanced than those of control groups.Conclu- sion:JEP has an effectively action on the differentiation of osteoblast-like cells in vitro.更多还原
