【摘要】 目的:探讨健骨二仙丸含药血清调节成骨细胞增殖、分化以及防止成骨细胞凋亡的机制。方法:采用原代成骨细胞培养,定量 RT-RCR 和 Northern 印迹杂交等方法观察成骨细胞胰岛素样生长因子(in-sulin-like growth factor-1)IGF-ImRNA 的表达。结果:健骨二仙丸能显著增加成骨细胞 IGF-ImRNA 的表达。结论:健骨二仙丸促进成骨细胞 IGF-ImRNA 的表达,从而进一步促进成骨细胞的增殖、分化,改善成骨细胞功能,调节骨重建偶联。表明健骨二仙丸不仅能增加成骨细胞胶原蛋白的合成,而且能促进非胶原蛋白地表达。促进骨的形成是健骨二仙丸防治骨质疏松症的主要机制。更多还原

【Abstract】 Objective:To explore the mechanisms of Jiangu Er’xian Pill (JEP) drug-containing serum on proliferation and differentiation of osteoblast,and on the prevention of osteoblast apoptosis.Methods:Human osteoblast-like cells were isolated and cuhured according to references.Quantitative reverse transcript polymerase chain reactions and Northern blot hybridization were applied to detect the in- sulin-like growth factor-Ⅰ (IGF-Ⅰ) mRNA in the cells.Results:JEP have a positive effect on the expression of IGF-Ⅰ mRNA in human osteoblast-like cells.Conclusion:JEP increases IGF-Ⅰ mRNA expression activates the human osteoblast-like cells prolifera- tion,differentiation,improves the function of these cells and modulates bone-remodeling coupling.These indicate that JEP not only have a positive action in increasing the synthesizing of collagen of human osteoblast-like cells,but also the constructing of non-collage- nous proteins and facilitating the formation of bone,and these may be the mechanism of JEP in preventing osteoporosis.更多还原
