左旋四氢巴马汀在大鼠急性脑缺血再灌注时对凋亡相关基因蛋白表达的影响
Effect of L-Tetrahydropalmatine on Expression of Gene Protein Related to Neuronal Apoptosis after Cerebral Ischemia/Reperfusion in Rats
【摘要】 目的:观察左旋四氢巴马汀(L-tetrahydropalmatine,L-THP)在脑缺血再灌注时对凋亡相关基因蛋白表达的影响。方法:在建立大鼠急性全脑缺血再灌注模型基础上,采用免疫组织化学法、原位末端标记法,分别检测假手术组、脑缺血再灌注组及 L-THP 治疗组:海马 CAl 区 Caspase-3、Fas、c-Fos、c-Myc、P53阳性细胞及凋亡细胞数。结果:脑缺血再灌注时,Caspase-3、Fas、c-Fos、C-Myc、P-53阳性细胞及细胞凋亡数均增加,与假手术组比较差异有显著性(P<0.01),L-THP 可减少这几项参数,L-THP 治疗组与脑缺血再灌注组比较差异亦有显著性(P<0.05,P<0.01)。结论:在脑缺血再灌注细胞凋亡过程中,L-THP 可通过抑制上述凋亡相关基因蛋白的表达,减少神经元凋亡,对脑缺血损伤起保护作用。
【Abstract】 Objective:To study the effects of L-tetrahydropalmatine(L-THP) on the expression of gene protein related to neuronal apopto- sis after cerebral ischemia/reperfusion in rats.Methods:Pulsinelli’s 4 blood vessels occlusion model was established to induce cerebral ischemia/reperfusion.Three groups were divided:sham-operation group(SO),ischemia/reperfusion group (IR) and L-THP treated group(THP).Changes in protein expression of Caspase-3,Fas,c-Fos,c-Myc,P53 were evaluated by immunohistochemical method. The number of neuronal apoptosis was assessed by in situ end labeling.Results:The positive cells of Caspase-3,Fas,c-Fos,c-Myc, P53 in the IR group was much more than those in the SO group ((P<0.01 ).Compared with IR group,the positive cells were much reduced than those in THP group (P<0.05,P<0.01).Conclusion.After cerebral ischemia/reperfusion,L-THP can protect neu- rons from ischemia and injury by suppressing the protein expression and blocking the neuronal apoptosis.
