半边莲不同组分对大鼠动脉内皮细胞内皮素合成及中膜平滑肌细胞增殖的影响

【摘要】 目的:探讨半边莲抗动脉粥样硬化作用的有效成分。方法:提取半边莲两种不同组分(A001,B001)用于高脂血症大鼠,观察其对动脉内皮细胞内皮素(ET)-1和内皮源型一氧化氮合酶(eNOS)代谢以及主动脉中层平滑肌细胞增殖的影响。结果:高脂大鼠应用半边莲 B001组分60天后,增高的血脂不降低,而血浆 ET-1浓度较高脂对照组明显降低(P<0.05);动脉内皮细胞及动脉壁中层平滑肌细胞 ET-1阳性细胞率和中膜厚度显著低于高脂对照组(P<0.05);血浆 eNOS 浓度则显著高于高脂对照组(P<0.05)。应用半边莲 A001组分的高脂大鼠血脂、血浆 ET-1、eNOS 浓度、动脉内皮细胞及动脉壁中层平滑肌细胞 ET-1阳性细胞率均无明显变化。结论:半边莲 B001使高脂血症大鼠内皮细胞、动脉平滑肌细胞合成及释放 ET-1减少,并使血浆 eNOS 含量增加,具有缓解高脂血症对血管内皮的持续损伤及抑制动脉平滑肌细胞增殖的作用。

【Abstract】 Objective:To explore the active principle of Scutellaria barbata (SB) in treating atherosclerosis.Methods:Two ingredients of SB,A001 and B001 were used in hyperlipidemic rats for 60 days to observe their effect on the endothelin-I (ET-1) of arterial en- dothelial cells and the metabolism of endothelial nitric oxide synthase (eNOS),the positive rate of ET-1 and aortic media smooth muscle cell(SMC)proliferation and the thickness of aortic media.Results:The plasma ET-1 concentration,the positive rate of ET- 1 in arterial endothelium and media SMC,and the thickness of aorta media reduced significantly (P<0.05),the blood eNOS in- creased significantly (P<0.05)in the B001 group compared with hyperlipidemia group.The morphological injury of the arterial en- dothelium was also alleviated obviously in B001 group.No significant changes were found in A001 group.Conclusion:The B001 is the active principle in SB,which can protect the arterial endothelium and impede the proliferation of VSM.
