氧化损伤类阿尔茨海默病大鼠模型的建立及调心方的作用
Establishment of Analogous Oxidative Damaged Alzheimer′s Disease Rat Model and Effect of Tiaoxin Recipe on It
【摘要】 目的 :建立一种简便、经济、实用的氧化损伤类阿尔茨海默病 (Alzheimer′sdisease ,AD)大鼠模型 ,探讨调心方对该模型大鼠空间学习记忆能力和 β 淀粉样蛋白质 (β amyloidprotein ,Aβ)沉积的影响。方法 :采用二羟延胡索酸 (DHF)加三氯化铁 -二磷酸腺苷 (FeCl3 ADP)左侧脑室注射的方法 ,建立氧化损伤型类AD大鼠模型 ;Morris水迷宫法观察大鼠空间学习记忆能力 ;免疫组化法观察大脑皮层Aβ沉积及调心方的作用。结果 :与对照组比较 ,模型鼠空间学习记忆能力显著下降 ,皮层Aβ广泛沉积 ,皮层Aβ阳性细胞数和免疫组化着色平均光密度显著增高 ;调心方对模型鼠上述指标变化有明显的改善作用。结论 :氧化损伤模型既能较好地表达AD的临床特征 (近期记忆损害 ) ,又能部分反应AD的病理变化 (Aβ沉积 ) ,是一经济实用的氧化损伤类AD大鼠模型 ;调心方具有改善氧化损伤类AD大鼠空间学习记忆障碍、降低Aβ沉积的作用。
【Abstract】 Objective:To establish a convenient, economical and practical analogous oxidative damaged Alzheimer′s disease rat model (AD model) for exploring the effect of Tiaoxin Recipe (TXR) on the spatial memory capacity and β-amyloid protein (Aβ) deposition in the model. Methods:The AD model was established by left ventricular injection of DHF-FeCl 3-ADP. Spatial memory and learning capacity of the model rat was observed by Morris water maze method, Aβ deposition in its cerebral cortex was observed by immunohistochemistry, and the effect of TXR was analysed. Results:Compared with the normal group, the spatial memory capacity in the model group was obviously decreased, with Aβ widely deposited in cortex, immunohistochemical examination showed that the number of Aβ positive cells and their mean optic density significantly increased. TXR displayed significantly improving effect on the above-mentioned changes. Conclusion:The oxidative damaged model could not only express the clinical characteristics (short-term memory impairment), but also partially reflex the pathological changes (Aβ deposition )of AD, is an economical and practical analogous AD model. TXR has the effects of improving spatial memory impairment and lowering Aβ deposition in the AD model rats.
