EGF及其受体flt对荷瘤鼠喉癌细胞增殖及凋亡的影响

【摘要】 目的 探讨VEGF及其受体flt对荷瘤鼠喉癌细胞增殖及凋亡的影响。方法 首先建立荷瘤 (HEP 2喉癌细胞 )裸鼠动物模型 ,将抗VEGF抗体及抗VEGF受体flt抗体分别注射荷瘤鼠体内 ,通过活体标记BrdU的方法 ,观察抗VEGF抗体及抗flt抗体对喉癌HEP 2细胞增殖的影响 ;用Tunel法观察对喉癌细胞凋亡的影响。结果 抗VEGF及抗flt抗体组HEP 2细胞BrdU标记阳性率显著小于对照组 (P <0 .0 5 ) ,细胞凋亡指数则显著高于对照组 (P <0 .0 5 )。结论 以抗体中和VEGF及封闭VEGF的结合位点均可抑制喉癌细胞的增殖 ,促进喉癌细胞凋亡。VEGF与喉癌细胞的生长密切相关。

【Abstract】 Objective To observe the effect of VEGF and its receptor on the growth and apoptosis of the laryngeal HEP-2 cancer Hep-2 cell line. Methods Laryngeal HEP-2 cancer xenograft was established in nude mice, and then treated the tumor-bearing mice with anti-VEGF antibody and its receptor flt antibody, and observed their effects on the growth of HEP-2 cell by labeling BrdU in vivo and on the apoptosis by in situ end-labeling method. Results The number of BrdU ladeled cells in the groups treated with anti-VEGF antibody and anti-flt antibody was significantly lower than that of control group(P<0.05) while the apoptosis index (AI) was significantly higher (P<0.05) There were no differences between the groups treated with anti-VEGF antibody and anti-flt antibody. Conclusion Both neutralizing the VEGF with antibody and blocking the flt receptor could inhibit the growth and promote the apoptosis of laryngeal cancer cell. VEGF seems to be highly associated with the growth of laryngeal cancer cells.
