平肝化浊合剂对脑梗塞大鼠神经行为学与脑组织缺血损伤的影响

【摘要】 目的 探讨平肝化浊合剂对脑梗塞大鼠神经行为学与脑组织缺血损伤的影响。方法应用10~12月龄雄性SD大鼠,随机分为假手术组(A)、造模组(B)、平肝化浊合剂组(C)、尼莫地平组(D),B、C、D组用同种大鼠无菌自然干燥的血凝块制作多发性脑梗塞模型。A、B组灌服蒸馏水,C组灌服平肝化浊合剂(25.40 g生药/kg),D组灌服尼莫地平片溶液(10.80 mg/kg),每天给药1次,连续2周。给药结束后,观察各组大鼠死亡率和用药前后的神经行为学分级,并取脑组织作病理组织学检查。结果 B组在用药1周后的死亡率明显升高(P<0.05);用药14 d后C组神经行为学分级明显优于B组(P<0.05);病理组织学检查发现,C组和D组大鼠脑组织中神经细胞光镜下的缺血性变化及其超微结构改变均较B组轻微。结论 平肝化浊合剂具有保护大鼠栓塞后脑组织的缺血损伤、改善肢体运动和协调功能、降低脑梗塞大鼠的早期死亡率的作用。

【Abstract】 Objective To assess the effects of Pigan Huazhuo Mixture (PHM) on neurogenic behavior and the cerebral ischemic damage in rats with cerebral infarct. Methods Male SD rats at 10 to 12 months old were randomly allocated to four groups: mimic operation group (group A), model group (group B), PHM group (group C) and nimodipine group (group D). Group B, C and D were injected with naturally-dried aseptic blood clots from the same rat species to establish models of multi-cerebral infarction. Group A and B were fed with distilled water (20 mL/kg), group C with PHM (25.40 g/kg) and group D with nimodipine (10.80 mg/kg), per time each day. After two-week treatment, the death rate and the neurogenic behavior gradating were detected and brain tissues were observed with pathological methods. Results One week after treatment, the death rate in group B was increased obviously(P<0.05). After two-week treatment, the neurogenic behavior gradating in group C was superior to that of group B (P<0.05). Histopathological results showed that the histomorphological and uhrastructural changes of neurons in group C and D were mild as compared with those in group B(P<0.05). Conclusion PHM can prevent brain tissue in rats with cerebral infarct from ischemic damage, promote the motor and coordination function of body and decrease the early death rate.
