脂多糖诱导大鼠肝星状细胞增殖与提高其胶原表达水平的细胞模型研究
Development of Cellular Model of Rat HSC Activation Stimulated by LPS
【摘要】 目的:建立较为全面反映肝星状细胞(hepatic stellate cell,HSC)活化效应的细胞模型。方法:应用脂多糖(lipopolysccharide,LPS)刺激体外培养的大鼠HSC。噻唑蓝(MTT)法测定HSC增殖率。免疫组化法显示HSC表达其Ⅰ、Ⅲ型胶原的表达水平,并进行图象分析。结果:LPS明显刺激HSC的增殖并提高Ⅰ、Ⅲ型胶原表达水平,与对照组比较差异有非常显著性意义(P<0.01)。结论:LPS诱导HSC活化模型能够相对全面地反映HSC活化的效应。
【Abstract】 Objective: Development of cellular model 1 of HSC activation. Methods: Isolated rat HSC cultured in vitro was stimulated with lipopolysaccharide (LPS). Proliferation of rat HSC was detected with MTT technique. Expression levels of collagen I and collagen Ⅲ in rat HSC were showed with immunochemistry technique and analyzed with image analysis system. Results: LPS significantly promoted proliferation elevated expression levels of collagen I and collagen M in rat HSC. Conclusion: Model of rat HSC activation stimulated with LPS can reflect more intact HSC activation effects.
