大鼠脑缺血再灌注损伤后血清中TNF-α变化及雷公藤多甙的影响
CHANGES OF SERUM TNF-α AFTER FOCAL CEREBRAL ISCHEMIC-REPERFUSION INJURY IN RATS AND EFFECTS OF MULTI-GLYCOSIDES OF TWHF ON THEM
【摘要】 目的 :研究肿瘤坏死因子 -α(TNF -α)在脑缺血再灌注损伤后的动态变化及雷公藤多甙 (GTW )对其影响。方法 :用线栓法建立大鼠脑缺血再灌注损伤模型 ,用放免法测定血清中TNF -α含量变化 ,灌胃法给药。结果 :大鼠脑缺血 1h再灌注 1h ,血清中TNF -α无变化 ,3h升高 ,6h达高峰 ,12h后下降 ,2 4h仍高于假手术组。GTW能抑制TNF -α水平升高 ,生理盐水治疗组与缺血再灌注组水平相同。结论 :大鼠脑缺血再灌注损伤后 ,TNF -α合成、分泌增加 ,GTW能降低血清中TNF -α含量。
【Abstract】 Objective:To study the changes of serum tumor necrosis factor-α(TNF- α ) after cerebral ischemic-reperfusion injury in rats and the effects of multi-glycosides of TWHF (GTW) on them.Methods:Rats’ cerebral ischemia-reperfusion injury model was established with suture method. Serum TNF- α contents were determined by radioimmunoassay (RIA) method.GTW was administered to rats oarally via gastric tube.Results:After 1 hour ischeamia followed by 1 hour reperfusion, TNF-α level in rats’ serum didn’t show great change. After 3 hours reperfusion, TNF-α level began to increase and reached peak at 6 hours. 12 hours later, TNF-α level began to decrease gradually, but was still much higher than that of sham group at 24 hours. GTW could inhibit the increase of TNF-α, but no difference was found between the normal saline treated group and the ischeamic-reperfusion group.Conclusion:After cerebral ischemic-reperfusion injury, the synthesis and secret of TNF-α were increased. GTW could reduce serum TNF-α level.
